Callus Induction and Regeneration from Germinating Mature Embryos of Wheat (Triticum aestivum L.)

نویسندگان

چکیده

A high-performance protocol for callus induction was devised using germinating mature embryos of two local wheat (Triticum aestivum L.) landraces as explant. The results showed that development from rapid starting one day after culture with an rate 20 to 25% higher than those soaked embryos. In addition, the mean growth developed 60 70% cultured This study also demonstrated a frequency green spots formation(48 56%)on derived compared their counterpart (24 28%), suggesting better differentiation potential cultures embryo. These findings indicate embryo is more suitable explant and regeneration commonly employed culture.

Regeneration of Freezing-Tolerant Spring Wheat (Triticum aestivum L.) Plants from Cryoselected Callus.

A cryoselection protocol has been developed that provides freezing-tolerant callus that, in turn, can regenerate plants with enhanced cold hardiness. Tolerant calli were selected from spring wheat (Triticum aestivum L.) callus by immersion in liquid nitrogen without addition of cryoprotectants. Less than 15% of the calli survived the initial challenge, whereas 30 to 40% of previously selected c...

Identification and Comparative Analysis of microRNA in Wheat (Triticum aestivum L.) Callus Derived from Mature and Immature Embryos during In vitro Culture

Feasible and efficient tissue culture plays an important role in plant genetic engineering. Wheat (Triticum aestivum L.) immature embryos (IMEs) are preferred for tissue culture to mature embryos (MEs) because IMEs easily generate embryogenic callus, producing large number of plants. The molecular mechanisms of regulation and the biological pathways involved in embryogenic callus formation in w...

The wheat (Triticum aestivum L.) leaf proteome.

The wheat leaf proteome was mapped and partially characterized to function as a comparative template for future wheat research. In total, 404 proteins were visualized, and 277 of these were selected for analysis based on reproducibility and relative quantity. Using a combination of protein and expressed sequence tag database searching, 142 proteins were putatively identified with an identificat...

Callus Induction and Somatic Embryogenesis from Maize Mature Embryos (zea Mays L.)

Ten maize lines prevalently used by breeders in the maize program were screened for their ability to form type II callus. Mature seeds were used as explant for in vitro culturing. Inbred lines showed high embryogenic response and formed friable, type II calli. And also exhibited successful plant regeneration. Various culture media compositions based on Murashige and Skoog (1962) salts thus inve...

Agrobacterium-mediated transformation of mature embryos of Triticum aestivum and Triticum durum

Plant regeneration studies in cereals have been undertaken in immature embryos, scutellum and also in immature inflorescence tissue. The wheat mature embryos can also be employed for callusing and regeneration, as they are available throughout the year and have presently been employed for transformation studies. An efficient and reproducible method for Agrobacterium-mediated transformation of m...
